چکیده
This study was designed with the aim of exploring efficacy RNA polymerase beta subunit, rpoB gene analysis for identification streptomycetes. A characteristic marine environment distinguished by their content streptomycetes were targeted, and two organisms genus Streptomyces isolated in Saudi Arabia. The spp., named isolates EH1 EH2 subjected to regular phenotypic characterization including morphological physiological studies, analyses cells hydrolysates, detecting antibacterial activities pocks formation ability. These showed inhibitory activity gram-positive bacteria Staphylococcus aureus Bacillus subtilis. gave positive results test, which indicated that both must belong Streptomyces. Molecular accomplished through beta-subunit (rpoB) assist species identification. target fragments 352 bp amplified detected using agarose gel electrophoresis, indicating they product isolate sequenced 296 continuous sequence determined (GenBank accession MK569762.1). By matching obtained DNA databases, found very similar labedae. concluded possibility molecular subunit correctly identify level.

Identification of Leptospira serovars by RFLP of the RNA polymerase beta subunit gene (rpoB)
Leptospires are usually classified by methods based on DNA-DNA hybridization and the conventional cross-agglutination absorption test, which uses polyclonal antibodies against lipopolysaccharides. In this study, the amplification of the rpoB gene, which encodes the beta-subunit of RNA polymerase, was used as an alternative tool to identify Leptospira. DNA extracts from sixty-eight serovars were...
متن کاملIdentification of mutations in the rpoB encoding the RNA polymerase beta subunit in rifampicine-resistant Mycobacterium tuberculosis strains from Iran.
The aim of this study was to investigate the frequency, location and type of rpoB mutations in Mycobacterium tuberculosis isolated from patients in Iran. 91 sputum were collected from suspected tuberculosis patients, 34 Rif-r isolates (87%) were identified as M. tuberculosis. Polymerase chain reaction (PCR) amplification and DNA sequencing methods were performed. 411 bp fragments of rpoB gene w...
متن کاملMolecular Identification of Some Schistosoma mansoni Isolates in Saudi Arabia
In this paper, nine geographical isolates of Schistosoma mansoni (three from Egypt, three from Saudi Arabia and 3 from Puerto Rico) were studied at the genotype level by RAPD analysis with two arbitrary primers. The genetic distance was measured by the percentage of unshared bands. The RAPD results showed that the Egyptian strains were closely related to Saudi strains but Puerto Rico strains cl...
متن کاملThe Euglena gracilis chloroplast rpoB gene. Novel gene organization and transcription of the RNA polymerase subunit operon.
The rpoB gene coding for a beta-like subunit of the chloroplast DNA-dependent RNA polymerase has been located on the chloroplast genome of Euglena gracilis distal to the rrnC ribosomal RNA operon. We have determined 5760 base-pairs of DNA sequence, including 97 bp of the 5S rRNA gene, an intergenic spacer of 1264 bp, the rpoB gene of 4249 bp, 84 bp spacer and 67 bp of the rpoC1 gene. The rpoB g...
متن کاملEnhancement of RNA Interference Effect in P19 EC Cells by an RNA-dependent RNA Polymerase
Background: RNA interference (RNAi) is a phenomenon uses double-stranded RNA (dsRNA) to specifically inhibit gene expression. The non-specific silencing caused by interferon response to dsRNA in mammalian cells limits the potential of utilizing RNAi to study gene function. Duplexes of 21-nucleotide short interfering dsRNA (siRNA) inhibit gene expression by RNAi. In some organisms, siRNA can als...
